Overview

Adoport (also known as Adport) is an immediate-release oral formulation of tacrolimus, a potent calcineurin inhibitor developed and marketed by Sandoz. As a small molecule immunosuppressant, tacrolimus works by binding to the intracellular protein FKBP12; the resulting complex then binds to and inhibits the phosphatase activity of calcineurin. This inhibition prevents the dephosphorylation and subsequent nuclear translocation of the nuclear factor of activated T-cells (NFAT), thereby blocking the transcription of genes for cytokines such as interleukin-2 (IL-2) that are essential for T-lymphocyte activation and proliferation. Adoport is primarily indicated for the prophylaxis of organ rejection in kidney, liver, and heart transplant recipients, as well as for the treatment of allograft rejection episodes resistant to other immunosuppressive regimens. It is designed for twice-daily administration and is bioequivalent to the reference product Prograf.
