ADoTope is a first-in-class dual-payload bi-epitope antibody-drug conjugate (ADC) platform developed by Medicovestor Inc. It is designed to target folate receptor alpha (FRα), which is highly expressed in platinum-resistant epithelial ovarian cancer. The platform utilizes a pair of fully human, high-affinity IgG1 monoclonal antibodies (Clone 65 and Clone 91), each recognizing distinct, non-overlapping epitopes on FRα. These antibodies are conjugated via enzyme-sensitive linkers to two different cytotoxic payloads: gemcitabine, a nucleotide analog, and exatecan, a topoisomerase I inhibitor. The mechanism of action involves the targeted delivery of these cytotoxic drugs to FRα-expressing cancer cells, followed by receptor-mediated internalization and intracellular release of the payloads, leading to cell death. Additionally, the naked antibodies contribute to antitumor activity through antibody-dependent cellular cytotoxicity (ADCC). The ADoTope platform aims to enhance cytotoxicity and reduce the risk of resistance by increasing antigen copy number and enabling dual-payload conjugation. The specific ADC developed using this platform, MC003, is currently in the IND-enabling stage for platinum-resistant epithelial ovarian cancer.
