ADoTope-MC003 is a preclinical **folate receptor alpha-targeted dual-payload bi-epitope antibody-drug conjugate** being developed for platinum-resistant epithelial ovarian cancer. The asset is built on the ADoTope platform and comprises two fully human high-affinity IgG1 antibodies, Clone 65 and Clone 91, that bind distinct non-overlapping epitopes on folate receptor alpha, functionally increasing target engagement on FRα-expressing tumor cells. In reported preclinical studies, Clone 65 was conjugated to gemcitabine and Clone 91 to exatecan through enzyme-sensitive linkers, creating a fixed-combination chemoimmunotherapy-style ADC designed to deliver mechanistically distinct payloads while also retaining antibody-mediated effector activity such as ADCC. Public conference abstracts describe enhanced in vitro potency versus corresponding single-payload comparators and activity in platinum-resistant ovarian cancer models, supporting further development in FRα-positive ovarian cancer.
