Overview

ADP-A2AFP is an investigational autologous T-cell therapy developed by Adaptimmune Therapeutics for the treatment of advanced cancers expressing alpha-fetoprotein (AFP). It consists of SPEAR (Specific Peptide Enhanced Affinity Receptor) T-cells engineered to express an affinity-enhanced T-cell receptor (TCR) that specifically targets AFP peptides presented by HLA-A*02:01. The therapy works by collecting T cells from patients, genetically modifying them in the laboratory to express the AFP-targeting TCR, and then infusing them back into the patient following lymphodepletion chemotherapy with fludarabine and cyclophosphamide. The primary target indication is hepatocellular carcinoma (HCC), though it has also been investigated in other AFP-expressing tumors including gastric hepatoid carcinoma, cholangiocarcinoma, and neuroendocrine tumors.
