Overview

AdPGp19 is a p53-responsive adenoviral vector designed to express the p19ARF protein, which is the murine homolog of the human p14ARF tumor suppressor. Developed by researchers at the University of Sao Paulo, the vector utilizes the AdPG system where transgene expression is driven by a p53-responsive promoter, creating a targeted expression profile in p53-active environments. It is primarily investigated for the treatment of malignant melanoma, aiming to restore the p53/ARF pathway to induce senescence and apoptosis in cancer cells. Preclinical studies have evaluated its efficacy both as a single agent and in combination with other gene therapies, such as those expressing interferon-beta (IFNβ), to enhance anti-tumor responses and inhibit tumor proliferation.
