ADPO-002 is a first-in-class, preclinical pharmaceutical drug developed by Adipo Therapeutics for the treatment of obesity and related metabolic disorders. It is a Notch signaling pathway inhibitor, specifically acting as a γ-secretase inhibitor, delivered via proprietary biodegradable poly(lactide-co-glycolide) (PLGA) nanoparticles (ADPO-002NP). The drug is injected directly into adipose tissue to provide localized delivery and sustained intracellular release. Its mechanism of action involves inhibiting Notch signaling in white adipose tissue, which induces browning (beiging) of fat through upregulation of key genes such as PRDM16 and PGC1a. This process increases mitochondrial biogenesis and energy expenditure by converting energy-storing white fat into metabolically active brown/beige fat. Preclinical studies have shown that this leads to systemic effects including weight loss, improved blood glucose control, and decreased lipid levels without affecting calorie intake. ADPO-002 remains at the injection site with minimal systemic exposure, reducing potential off-target side effects.
