aDR5-expressing CAR-T cells are an engineered cell therapy designed to enhance the efficacy of CAR-T therapy against solid tumors. This therapy involves CAR-T cells that are modified to express a membrane-bound DR5 agonist (aDR5) which lacks intracellular signaling capabilities. The aDR5 component induces apoptosis in DR5-sensitive tumor cells, myeloid-derived suppressor cells, and tumor-associated macrophages, independently of CAR engagement. This mechanism aims to overcome the immunosuppressive tumor microenvironment often found in solid tumors, thereby improving tumor control without exacerbating CAR-T cytokine responses or relying solely on T cell fitness.
