Overview

AdRGD-FKN is a gene therapy investigational product comprising a fiber-mutant adenoviral vector engineered to deliver the gene encoding murine fractalkine (FKN, also known as CX3CL1) into target tissues, primarily for cancer immunotherapy research. The vector is modified by the insertion of an RGD motif into the fiber protein to enhance transduction efficiency, particularly in cells that lack the Coxsackie and adenovirus receptor. Upon intratumoral injection, AdRGD-FKN causes localized expression of FKN, leading to infiltration of T cells and NK cells within the tumor. However, as a monotherapy, it does not induce a notable anti-tumor effect, likely due to limited activation of infiltrating immune cells. Its primary use has been studied in combination with other cytokine gene therapies, such as AdRGD-IL-12, where a synergistic effect results in potent tumor regression and long-term immune protection in preclinical animal cancer models. AdRGD-FKN is an experimental tool, not a commercially available pharmaceutical, and is used in preclinical research settings.[1]
