AdRGD-IL-12 is an investigational gene therapy product consisting of an **adenoviral vector encoding interleukin-12 (IL-12)**, engineered with an RGD motif to enhance tumor targeting. This agent is designed for **intratumoral administration** to induce **local expression of IL-12**, a potent pro-inflammatory cytokine known to stimulate both innate and adaptive antitumor immune responses, chiefly through activation of cytotoxic T and NK cells. Preclinical studies showed that AdRGD-IL-12, alone or in combination with other targeted vectors (such as AdRGD-FKN), can induce significant tumor regression, enhance **immune cell (T and NK) infiltration**, and promote lasting antitumor immunity in mouse cancer models. The RGD modification facilitates preferential targeting of αv integrin-expressing tumor cells, aiming for increased intratumoral IL-12 expression and reduced systemic toxicity. Primary research and experiments involving this vector have demonstrated robust induction of local immune activation, perforin-positive cell infiltration, and, when combined with other immune-modulating vectors, enhanced efficacy for tumor clearance and immunological memory[3].
