This is a combination chemotherapy regimen consisting of adriamycin (doxorubicin) and amsacrine (m-AMSA). Both are cytotoxic small molecules used as antineoplastic agents. Doxorubicin is an anthracycline antibiotic that exerts its anticancer effects primarily by intercalating into DNA and inhibiting topoisomerase II, leading to DNA damage and generation of reactive oxygen species. Amsacrine is an acridine derivative that also acts as a topoisomerase II poison by stabilizing the enzyme-DNA cleavage complex and preventing religation of DNA strands. This combination has been studied in advanced breast cancer and hematologic malignancies but has limited efficacy in solid tumors like breast cancer[1][5]. The primary indications for these drugs individually are various cancers including leukemias (especially acute leukemias), lymphomas, sarcomas, and breast cancer[2][4][7].
