Overview

This is a combination chemotherapy regimen consisting of three cytotoxic agents: - Adriamycin (also known as doxorubicin), an anthracycline antibiotic that intercalates DNA and inhibits topoisomerase II, leading to inhibition of DNA replication and cell death. - Cyclophosphamide, an alkylating agent that crosslinks DNA strands, preventing cell division and leading to apoptosis. - Melphalan, another alkylating agent that also crosslinks DNA and disrupts cancer cell proliferation. The combination has been shown in preclinical models (such as L1210 murine leukemia) to be highly synergistic, resulting in significantly improved survival compared to the use of each drug alone at optimal doses. The regimen substantially inhibits tumor cell DNA synthesis while having less impact on sensitive host tissues. This combination is used for its enhanced antitumor efficacy due to the complementary mechanisms of action[1].
