Overview

Adriforant is an oral, small molecule drug that acts as a selective antagonist of the histamine H4 receptor. It was originally developed by Pfizer and later by Novartis (through its subsidiary Ziarco Pharma) for the treatment of inflammatory skin conditions such as atopic dermatitis and psoriasis. The drug demonstrated preclinical efficacy in reducing itch and inflammation via functional inhibition of the histamine H4 receptor, but clinical trials in humans did not meet efficacy endpoints, leading to discontinuation of its development for these indications[3][6][7][8]. Adriforant reached up to phase II clinical trials for atopic dermatitis and psoriasis, with additional early-stage investigation in asthma[6][7].
