Overview

Adrogolide (DAS-431; ABT-431) is a chemically stable prodrug that is rapidly converted in plasma to A-86929, a full agonist at the dopamine D1 receptor. It is over 400 times more selective for dopamine D1 than D2 receptors. Adrogolide has demonstrated efficacy in improving behavioral disability and locomotor activity in animal models of Parkinson’s disease and shows antiparkinsonian effects equivalent to L-DOPA in patients with Parkinson’s disease, with potentially reduced risk of dyskinesia. It has also shown promise for cognitive impairment in the elderly and for reducing cocaine craving and subjective effects in cocaine-dependent individuals. The drug was developed as an intravenous formulation due to low oral bioavailability from high first-pass metabolism. Adverse events are generally mild-to-moderate and include injection site reactions, headache, nausea, dizziness, and postural hypotension[1][2][3][4][5].
