ADRX-0405

ADRX-0405 is a next-generation antibody-drug conjugate (ADC) developed by Adcentrx Therapeutics. It consists of a humanized IgG1 antibody targeting six-transmembrane epithelial antigen of the prostate 1 (STEAP1), a cell surface protein highly expressed in prostate cancer and certain other cancers, but with limited expression in normal tissues. The ADC uses Adcentrx’s proprietary i-Conjugation technology platform, which employs protease-cleavable linkers and stable conjugation chemistry to enhance payload delivery. The drug carries a novel topoisomerase inhibitor as its cytotoxic payload, conjugated at a drug-to-antibody ratio of eight (DAR 8). Preclinical studies have shown favorable pharmacokinetics, safety profile, and significant efficacy across multiple tumor models. ADRX-0405 is currently being evaluated in Phase 1 clinical trials for advanced solid tumors including metastatic castration-resistant prostate cancer, non-small cell lung cancer, small cell lung cancer, and gastrointestinal cancers[1][2][4][7].
