ADS024

ADS024 is an orally administered, non-engrafting, single-strain live biotherapeutic product (SS-LBP) consisting of the bacterium *Bacillus velezensis*. Originally known as ART24, it was developed by Artugen Therapeutics and is now being advanced by Adiso Therapeutics. The drug has a multimodal mechanism of action, which includes the degradation of *C. difficile* toxins, anti-inflammatory and anti-apoptotic effects, and protection of the gut barrier. [1, 2, 3, 4, 7, 10] More recently, it was identified as a strong and specific agonist of the lysophosphatidic acid receptor 3 (LPA3), a G-protein coupled receptor (GPCR) implicated in the modulation of neuroinflammation via the gut-brain-immune axis. [13, 14] ADS024 is in development for preventing the recurrence of *C. difficile* infection (for which it has received FDA Fast Track Designation) and for treating ulcerative colitis. [5] It has also shown efficacy in preclinical models of several neuroinflammatory diseases, including Parkinson's disease, multiple sclerosis, ALS, Huntington's disease, and chemotherapy-induced peripheral neuropathy.
