ADS032

ADS032 is a novel small molecule drug developed by Adiso Therapeutics that acts as a first-in-class dual inhibitor of the NLR family pyrin domain containing 1 (NLRP1) and NACHT LRR and PYD domains-containing protein 3 (NLRP3) inflammasomes, which are key components of the innate immune system involved in sensing pathogens and danger signals. By directly binding to both NLRP1 and NLRP3, ADS032 blocks their activation, thereby reducing secretion and maturation of pro-inflammatory cytokines such as IL-1β and TNF-α. This mechanism interrupts the downstream inflammatory cascade by inhibiting ASC speck formation, a hallmark of inflammasome activation. Preclinical studies have demonstrated that ADS032 reduces systemic and pulmonary inflammation in animal models—including protection against lethal influenza A virus challenge—and shows promise for treating respiratory and dermal inflammatory diseases as well as other conditions associated with chronic inflammasome activation[1][4][7].
