Overview

AdsFlk-1 is an adenoviral gene therapy vector designed to express a soluble fragment of the FLK-1 receptor, which is the murine homolog of the human vascular endothelial growth factor receptor-2 (VEGFR2). This soluble receptor (sFlk-1) acts as a dominant-negative decoy, binding to circulating vascular endothelial growth factor (VEGF) and preventing it from interacting with its membrane-bound receptors on endothelial cells. By sequestering VEGF, AdsFlk-1 effectively inhibits tumor-induced angiogenesis and has demonstrated significant antitumor efficacy in preclinical murine models of colorectal cancer (CRC), including metastatic disease. Research indicates that systemic administration of the vector can lead to a compensatory increase in circulating VEGF levels, although this does not necessarily correlate with a loss of antitumor treatment response.
