ADT-030

**ADT-030** is an investigational oral small molecule developed by ADT Pharmaceuticals as a dual **pan-RAS/β-catenin inhibitor** for treating **KRAS-driven cancers**, particularly **pancreatic ductal adenocarcinoma (PDAC)**. It potently inhibits proliferation of KRAS G12D- and G12C-mutant PDAC cells, induces apoptosis selectively in mutant cells, arrests the cell cycle at G2/M, and suppresses **MAPK/AKT** and **β-catenin** signaling pathways. In orthotopic mouse models of KRAS G12D PDAC, oral dosing (50-150 mg/kg) achieved up to 80% tumor growth inhibition with good tolerability and modulated the tumor immune microenvironment by depleting myeloid-derived suppressor cells while increasing CD8+ T cell and NK cell infiltration. The compound is formulated with an antacid for safety and is in IND-enabling studies.[1][5][7][9][13]
