Overview

ADU-1805 is a humanized immunoglobulin G2 (IgG2) monoclonal antibody that targets signal-regulatory protein alpha (SIRPα; CD172a), an immune checkpoint molecule expressed on myeloid cells. By binding to all known human SIRPα alleles and blocking the interaction between SIRPα and CD47, ADU-1805 disrupts the "don't eat me" signal used by cancer cells to evade phagocytosis. This blockade promotes macrophage-mediated phagocytosis of tumor cells and enhances neutrophil trogocytosis, with similar potency to anti-CD47 antibodies but potentially improved safety due to minimal binding to SIRPβ1 and restricted expression of SIRPα. Unlike CD47-targeting agents, it does not interfere with T-cell activation or cause frequent hematologic toxicities in preclinical models. The drug is being developed for the treatment of advanced solid tumors and refractory cancers[1][3][4][5][7].
