ADU-623 is a bivalent, live-attenuated Listeria monocytogenes-based therapeutic cancer vaccine engineered to express two tumor-associated antigens: EGFRvIII (epidermal growth factor receptor variant III) and NY-ESO-1 (cancer/testis antigen 1). The vaccine is designed to stimulate robust tumor-specific immune responses against cells expressing these antigens, which are commonly found in glioblastoma and other high-grade astrocytomas. In addition to eliciting adaptive immunity targeting EGFRvIII and NY-ESO-1, the live Listeria vector also induces potent innate immune activation that may contribute to anti-tumor effects even in the absence of these neoantigens. The primary clinical development has focused on patients with recurrent or treated WHO Grade III/IV astrocytic tumors following standard therapy[1][5][6][7][10].
