ADV-101

ADV-101 is a first-in-class antibody-drug conjugate (ADC) targeting the interleukin-1 receptor accessory protein (IL-1RAP), which is highly expressed in esophageal squamous cell carcinoma (ESCC) and other solid tumors. The drug consists of a humanized anti-IL-1RAP IgG1 monoclonal antibody conjugated via a cleavable peptide-based linker to a topoisomerase I inhibitor payload, with a drug-antibody ratio of 8. ADV-101 demonstrates sub-nanomolar binding affinity and effective internalization in ESCC cell lines, leading to potent cytotoxicity. In preclinical models, it achieved complete tumor responses in mice bearing ESCC tumors. The mechanism of action involves selective delivery of the cytotoxic payload to IL-1RAP-expressing cancer cells, resulting in targeted inhibition of topoisomerase I and disruption of DNA replication.
