Overview

AdV-lumc007dRbD is an oncolytic non-human primate adenovirus developed by the Oncolytic Viro-Immunotherapy (OVIT) consortium, which includes Leiden University Medical Center (LUMC), Erasmus Medical Center, and Radboud University Medical Center. The virus is engineered for tumor-selective replication, likely utilizing a deletion in the Rb-binding domain (indicated by the "dRbD" suffix) to restrict its activity to cancer cells with a dysfunctional Retinoblastoma (Rb) pathway. It is primarily being investigated for the treatment of urological malignancies, including bladder and prostate cancer. AdV-lumc007dRbD works by inducing direct oncolysis and promoting immunogenic cell death (ICD), which is characterized by the release of damage-associated molecular patterns (DAMPs) such as HSP90, Calreticulin, and HMGB1. This process facilitates the maturation and activation of dendritic cells, potentially triggering a systemic anti-tumor immune response.
