Advax-CpG55.2 is a combination adjuvant system developed by Vaxine Pty Ltd and evaluated by the National Institute of Allergy and Infectious Diseases (NIAID). It is composed of two active components: delta inulin (Advax), a plant-derived polysaccharide particle that targets DC-SIGN on dendritic cells, and CpG55.2, a synthetic B-class oligonucleotide with a phosphorothioate backbone that acts as a Toll-like receptor 9 (TLR9) agonist. This adjuvant is designed to enhance both humoral and cellular immune responses when formulated with vaccines. It has been primarily evaluated in Phase I clinical trials (NCT03945825) in combination with quadrivalent seasonal influenza vaccines, such as Fluzone (inactivated) and Flublok (recombinant), to improve antibody titers and neutralizing responses in healthy adults.
