**AdVCA0848** is a recombinant non-replicating adenovirus serotype 5 (Ad5) vector engineered to express the VCA0848 diguanylate cyclase gene from Vibrio cholerae, enabling in vivo synthesis of cyclic di-GMP (c-di-GMP), a potent STING pathway agonist. Developed through a partnership between Michigan State University and Venn Therapeutics, it targets integrin αvβ6 and αvβ8 to enhance delivery and activate innate immune responses, including dramatic induction of IFN-β and promotion of anti-tumor immunity. A single intra-tumoral injection demonstrated rapid inhibition of B16 melanoma tumor growth and improved survival in preclinical animal models compared to repeated cytokine treatments, positioning it as a novel immunotherapy for cancer.
