Overview

ADX‑10061 is a potent, selective small molecule antagonist of the dopamine D1 receptor. It was originally developed by Novo Nordisk and later by Addex Pharmaceuticals for potential use in treating nicotine dependence (smoking cessation), schizophrenia, sleep disorders, and other substance-related disorders. The drug acts as a competitive orthosteric antagonist at the dopamine D1 receptor, which is implicated in reward-seeking behaviors and cue-induced craving—mechanisms fundamental to addiction. Despite reaching Phase II clinical trials for several indications, development was discontinued after failing to demonstrate efficacy in smoking cessation trials. The compound has also been used as a research tool to study dopaminergic function in the brain[1][2][3][5][6].
