Overview

ADX-1615 is an oral pro-drug of ganetespib (ADX-1612), a clinical-stage small molecule inhibitor of Heat Shock Protein 90 (Hsp90). In-licensed by Aldeyra Therapeutics from Madrigal Pharmaceuticals in December 2016, ADX-1615 is being developed for the treatment of systemic inflammatory and autoimmune diseases. The drug acts as a chaperome (or epichaperome) inhibitor, targeting the protein folding machinery that is often hijacked or dysregulated in disease states to maintain the stability of pro-inflammatory client proteins. While its parent compound, ganetespib, was extensively investigated for oncology indications, Aldeyra's strategy involves repurposing the Hsp90 inhibition mechanism to modulate inflammatory pathways. The pro-drug design of ADX-1615 is specifically intended to enable oral administration, providing a more patient-friendly alternative to the intravenous formulations typically associated with this class of inhibitors.
