ADX71441 is an oral small molecule positive allosteric modulator of the GABA B receptor being developed by Addex Therapeutics for neurological and psychiatric conditions including anxiety, pain, spasticity, and substance use disorders; it enhances GABA B receptor function rather than directly activating the receptor, aiming to improve efficacy and tolerability versus orthosteric agonists such as baclofen.[5][7] Preclinical studies show brain penetration, oral bioavailability, anxiolytic-like effects, analgesic activity, muscle-relaxant qualities, and attenuation of alcohol and nicotine withdrawal behaviors; growth hormone modulation has been explored as a pharmacodynamic biomarker for target engagement.[7][10][3][2]
