ADX71743 is a potent, highly selective, and brain-penetrant small molecule **negative allosteric modulator (NAM)** of the **metabotropic glutamate receptor 7 (mGlu7)**. Developed by Addex Therapeutics, it serves as a critical research tool for investigating the physiological and therapeutic roles of mGlu7 in the central nervous system. The compound exhibits high selectivity for the mGlu7 subtype, with EC50 values in the nanomolar range (63 nM for human and 88 nM for mouse), and shows no significant activity at other mGlu receptor subtypes (mGlu3, mGlu4, mGlu5, mGlu6, and mGlu8). Preclinical studies have demonstrated that ADX71743 possesses anxiolytic-like and antipsychotic-like properties in various animal models, such as the marble burying and elevated plus maze tests, suggesting potential utility in treating anxiety disorders, post-traumatic stress disorder (PTSD), and schizophrenia. However, it has shown a lack of efficacy in models of depression.
