ADXS-HOT is a preclinical immunotherapy platform developed to target shared "hotspot" mutations in tumor driver genes and other tumor-associated antigens, including cancer-testes and oncofetal antigens. It leverages a proprietary Listeria monocytogenes (Lm) technology to deliver these antigens directly to the immune system, aiming to induce potent anti-tumor immunity by stimulating both innate and adaptive immune responses. The platform is designed for off-the-shelf use, potentially allowing treatment of all patients with a particular cancer type without the need for individualized diagnostic testing or sequencing. Constructs from the ADXS-HOT program are engineered as live attenuated Lm-based vectors that secrete antigen-adjuvant fusion proteins, which can generate high-avidity T cells against multiple targets[2].
