Overview

ADXS-NEO is a personalized, neoepitope-based cancer vaccine developed using live attenuated Listeria monocytogenes (Lm) engineered to deliver patient-specific tumor neoantigens. The therapy is designed to stimulate the immune system to recognize and attack cancer cells by presenting up to 40 unique mutation-derived antigens identified from sequencing each patient's tumor. Upon administration, the Lm vector delivers these neoantigen sequences into antigen-presenting cells, inducing a robust T-cell-mediated immune response against the patient’s specific cancer mutations. ADXS-NEO has been evaluated in Phase 1 clinical trials for various advanced or metastatic solid tumors including non-small cell lung cancer (NSCLC), microsatellite stable colorectal cancer (MSS-CRC), and squamous cell carcinoma of the head and neck. The drug was originally developed by Advaxis in collaboration with Amgen and Memorial Sloan Kettering Cancer Center; Advaxis later merged with Ayala Pharmaceuticals[1][4][5][6][7][8].
