ADXS31-142 is a live attenuated, bioengineered immunotherapy based on the bacterium *Listeria monocytogenes*. It is designed to secrete a fusion protein composed of a truncated listeriolysin O (tLLO) and human prostate-specific antigen (PSA), thereby stimulating an immune response against PSA-expressing tumor cells. The drug acts as a cancer vaccine and has been investigated both as monotherapy and in combination with pembrolizumab for the treatment of metastatic castration-resistant prostate cancer (mCRPC). Its mechanism involves activating the immune system to recognize and attack prostate cancer cells expressing PSA[1][2][3][4].
