AE-848 is a small molecule inhibitor of insulin-like growth factor 2 mRNA-binding protein 3 (IGF2BP3), a protein that functions as a reader of N6-methyladenosine (m6A) on RNA. By binding to IGF2BP3, AE-848 disrupts the stability of its target mRNAs, including c-MYC, VEGF, CDK2, CDK6, and STAT1, which are critical for tumor progression and chemoresistance. Beyond its direct effects on tumor epithelial cells, AE-848 modulates the tumor microenvironment by reducing c-MYC expression in M2 macrophages and tumor-associated macrophages (TAMs), while promoting the secretion of anti-tumor cytokines such as IFN-γ and TNF-α. Preclinical studies in ovarian cancer models have demonstrated that systemic administration of AE-848 significantly inhibits tumor growth and metastasis.
