A cancer vaccine composed of the AE37 peptide, which is a HER2/neu-derived epitope (amino acids 776–790) linked to the Ii-Key peptide, combined with granulocyte-macrophage colony-stimulating factor (GM-CSF). The vaccine is designed to stimulate an immune response against cancer cells expressing the HER2/neu antigen. The Ii-Key moiety enhances T-helper cell stimulation by increasing presentation of the HER2/neu antigen via MHC class II molecules. Upon vaccination, AE37 activates CD4+ helper T cells and can also induce memory T cells and cytotoxic CD8+ lymphocytes that target tumor cells. GM-CSF acts as an immunoadjuvant, further potentiating this immune response. This combination has been primarily investigated for preventing recurrence in breast cancer patients who are disease-free but at high risk due to node-positive or high-risk node-negative status[1][3][4][5][7].
