AE37 is a therapeutic cancer vaccine designed to stimulate the immune system against tumors expressing the HER2 protein. It is a hybrid peptide-based immunotherapy composed of two main components: a segment from the intracellular domain of the HER2 protein (AE36) and an immune-regulatory sequence called Ii-Key. The combination enhances antigen presentation and stimulates CD4+ T-helper cells, leading to both antibody and cellular immune responses against HER2-expressing cancer cells. The vaccine is administered with GM-CSF as an adjuvant to further boost immunogenicity. AE37 has been primarily developed for preventing recurrence in breast cancer patients who are clinically disease-free but have tumors overexpressing or under-expressing HER2; it has also been studied in prostate cancer and is being explored for other malignancies such as triple-negative breast cancer and bladder cancer[1][3][4][7][8].
