AEG-1 siRNA is a **small interfering RNA** designed to selectively inhibit the expression of *astrocyte elevated gene-1* (*AEG-1*, also known as *metadherin* or *MTDH*), a gene overexpressed in multiple cancers and implicated in tumor growth, metastasis, angiogenesis, chemoresistance, and epithelial-mesenchymal transition. AEG-1 promotes cancer progression partly by increasing expression of multidrug resistance proteins, supporting the PI3K/Akt and Wnt/β-catenin signaling pathways, and regulating the function of RNA-induced silencing complexes (RISC). Suppression of AEG-1 by siRNA has been shown in preclinical studies to decrease cancer cell proliferation, sensitize tumors to chemotherapeutics like doxorubicin and 5-fluorouracil, reduce metastasis, and reverse EMT phenotypes. Research is primarily at the preclinical stage, with most data from cellular and animal xenograft models[1][2][3][7].
