Overview

AEM-28-14 is a next-generation apolipoprotein E (apoE) mimetic peptide analog of AEM-28, engineered to more potently clear atherogenic apoB-containing lipoproteins via hepatic heparan sulfate proteoglycan–mediated pathways and related apoE receptors. Preclinical studies indicate approximately 400% greater cholesterol-lowering efficacy and several-fold increased drug exposure relative to AEM-28, with profound and rapid reductions in LDL cholesterol and VLDL/chylomicron remnants in hypercholesterolemic animal models.[4][7][8] Initially discovered at the University of Alabama at Birmingham and developed by Capstone Therapeutics, AEM-28-14 is being advanced as an intravenous biologic for severe refractory dyslipidemias, including homozygous familial hypercholesterolemia, under orphan-drug designation.[8][9][10]
