Overview

AERAS-422 is a recombinant BCG (rBCG) vaccine derived from the Danish 1331 strain of BCG. It was engineered to overexpress specific TB antigens (**Ag85A**, **Ag85B**, and **Rv3407**) and express a mutant perfringolysin (**PFO G137Q**) from *Clostridium perfringens*. The vaccine was designed for tuberculosis prevention, aiming to improve upon the standard BCG vaccine by using the PFO component to facilitate antigen entry into the cytosol and stimulate an MHC Class I-restricted immune response, potentially generating stronger and more persistent CD8+ T cell responses.
