Overview

Aerosolized vancomycin is an inhaled formulation of the glycopeptide antibiotic vancomycin, currently being investigated in Phase I clinical trials sponsored by the National Cancer Institute (NCI). It is specifically being evaluated for the treatment of advanced non-small cell lung cancer (NSCLC) as part of a combination regimen with aerosolized aztreonam and the immune checkpoint inhibitor pembrolizumab. The therapeutic rationale for this approach is based on the observation that patients with NSCLC often exhibit bacterial dysbiosis in their lungs, which may promote tumor growth and impair the efficacy of immunotherapy. By delivering vancomycin directly to the lungs via inhalation, researchers aim to selectively target Gram-positive bacteria within the lung microbiome to create a more favorable environment for pembrolizumab-mediated anti-tumor immune responses.
