Overview

Aerugen is an octovalent conjugate vaccine designed for the prevention of *Pseudomonas aeruginosa* infections, specifically in patients with cystic fibrosis. It consists of O-polysaccharides from eight different serotypes of *P. aeruginosa* conjugated to a detoxified recombinant form of exotoxin A (ETA). The vaccine is adjuvanted with aluminum hydroxide and works by stimulating the production of high-titer IgG antibodies against both the lipopolysaccharide (LPS) and exotoxin A of the bacteria, aiming to prevent initial colonization and subsequent chronic infection. Developed initially by Berna Biotech (Swiss Serum and Vaccine Institute) and later licensed to Crucell, Solvay, and Orphan Europe, the vaccine reached Phase III clinical trials but development was suspended in 2006 after failing to demonstrate efficacy in preventing colonization.
