AETX-R114 is a preclinical bispecific T cell engager (TCE) developed by Aethon Therapeutics using its proprietary HapImmune™ platform. It is designed to target synthetic neoantigens formed when the covalent KRAS G12C inhibitor sotorasib binds to mutant KRAS proteins within cancer cells. These drug-modified peptides are subsequently presented on the cell surface as peptide:MHC (p*MHC) complexes. AETX-R114 specifically recognizes these sotorasib-modified KRAS G12C peptides presented by HLA-A3 supertype alleles and simultaneously engages CD3 on T cells. This dual binding redirects T-cell cytotoxicity toward cancer cells, including those that have developed resistance to sotorasib, thereby overcoming targeted therapy resistance in KRAS G12C-mutant cancers.
