Overview

AETX-R302 is a preclinical monoclonal antibody developed by Aethon Therapeutics using its proprietary HapImmune™ platform. It is designed to target KRAS G12C-mutant cancers in a combination strategy with the covalent KRAS G12C inhibitor divarasib. The antibody specifically recognizes a unique neoantigen (hapten-pMHC) formed when divarasib covalently modifies the KRAS G12C protein, which is subsequently processed and presented on the cell surface by MHC molecules. AETX-R302 demonstrates high-affinity (KD ranging between 0.78 and 2.75 nM) and pan-HLA recognition across HLA-A*02 (A2 supertype) and HLA-A*03/A*11 (A3 supertype) alleles. By targeting these drug-induced neoantigens, AETX-R302 aims to direct the immune system to selectively eliminate cancer cells, including drug-resistant "persister" cells that may survive initial inhibitor treatment.
