Overview

AEV103 is a next-generation immunomodulatory imide drug (IMiD), chemically identified as 3,6'-dithiopomalidomide, being developed by AevisBio for neuroinflammation-driven neurodegenerative diseases such as Alzheimer’s and Parkinson’s disease. It binds strongly to cereblon, an E3 ubiquitin ligase used in targeted protein degradation, and exerts immunomodulatory effects by reducing production of the pro‑inflammatory cytokine tumor necrosis factor alpha (TNF-α) and attenuating microglial activation, thereby decreasing neuroinflammation, neuronal and synaptic loss, and cognitive decline in preclinical Alzheimer’s disease models, without altering amyloid beta levels.[1][4][10] Originally invented at the NIH and exclusively licensed to AevisBio, AEV103 represents a cereblon‑binding small‑molecule TNF‑α–lowering agent intended as a disease‑modifying therapy for neurodegenerative disorders driven by chronic inflammation.[1][4][10]
