Drug intelligence / Profile preview
AF-100S is a proprietary sub-micron formulation of fenofibrate developed by Afecta Pharmaceuticals. Fenofibrate is a small molecule agonist of the peroxisome proliferator-activated receptor alpha (PPARα), a nuclear receptor that plays a central role in lipid metabolism. Upon activation, PPARα increases the expression of genes involved in fatty acid oxidation and lipoprotein lipase activity, while decreasing the expression of apolipoprotein C-III. This results in a significant reduction in plasma triglyceride levels and an increase in high-density lipoprotein (HDL) cholesterol. The AF-100S formulation utilizes sub-micron particle technology to overcome the poor aqueous solubility of fenofibrate, thereby enhancing its dissolution rate and oral bioavailability. This improved pharmacokinetic profile is intended to provide more consistent therapeutic effects and potentially eliminate the requirement for administration with food. AF-100S has been investigated for the treatment of hypertriglyceridemia, mixed dyslipidemia, and more recently, for its potential to mitigate the inflammatory response in patients with COVID-19.
