AF-16 is a synthetic 16-amino acid peptide derived from the active functional fragment of the antisecretory factor (AF) protein. Developed by Lantmännen Medical AB (part of the Lantmännen group), it acts as an AF mimetic designed to modulate water and ion transport across cell membranes, thereby reducing intracranial pressure and brain edema. The drug is primarily being investigated for the treatment of cerebral oedema following traumatic brain injury (TBI). Preclinical studies have demonstrated that intranasal administration of AF-16 significantly reduces brain edema and improves cognitive outcomes in animal models. Clinical development has reached Phase 2, with an open-label, historical cohort-controlled study in Germany evaluating its safety and tolerability in adult patients with TBI-induced cerebral oedema. Related AF-based products, such as the egg yolk powder Salovum, are classified as foods for special medical purposes (FSMP).
