AF165-multiGS-Pep1 is a recombinant fusion protein consisting of the human recombinant anti-bone sialoprotein (BSP) antibody AF165 fused to the cell-penetrating peptide Pep1 via a multi-glycine-serine (GS) linker. Developed by the German Cancer Research Center and Immundiagnostik, this therapeutic candidate is designed to target intracellular bone sialoprotein (BSP) in cancer cells. While the parent antibody AF165 lacks intracellular uptake, the addition of the Pep1 cell-penetrating peptide enables efficient internalization into MDA-MB-231 breast cancer cells. Once inside, AF165-multiGS-Pep1 downregulates BSP levels, upregulates EGR1, ATF3, and ID2, and induces apoptosis, demonstrating potential for the treatment of breast cancer and associated bone metastases.
