AF165-multiGS-TAT is a recombinant fusion protein consisting of the human recombinant anti-bone sialoprotein (BSP) monoclonal antibody AF165 fused to the cell-penetrating peptide (CPP) TAT via a glycine-serine (GS) linker. Developed by the German Cancer Research Center (DKFZ) and Immundiagnostik, the fusion protein is designed to target bone sialoprotein (BSP), which is highly expressed in bone-metastatic malignancies such as breast cancer. Unlike the parent antibody AF165, which has limited cellular uptake, AF165-multiGS-TAT is efficiently internalized by cancer cells, where it downregulates BSP levels and upregulates pro-apoptotic proteins (EGR1, ATF3, and ID2), leading to apoptosis and inhibition of cell proliferation. It is being investigated preclinically for the treatment of bone-metastatic breast cancer.
