Overview

AF38469 is a small molecule inhibitor of **sortilin** (also known as SORT1), a multi-ligand receptor and intracellular chaperone belonging to the Vps10p family. Sortilin plays a critical role in various pathological processes, including lipid metabolism, inflammation, and vascular calcification. AF38469 specifically binds to the Vps10p domain of sortilin, thereby blocking its interaction with ligands and downstream signaling pathways. In preclinical studies, particularly in minipig models of calcific aortic valve disease (CAVD), AF38469 has demonstrated the ability to preserve endothelial integrity, reduce oxidative stress, and inhibit the activation of myofibroblast and osteoblast phenotypes that lead to valvular mineralization. Additionally, the compound has shown efficacy in reducing plasma cholesterol and triglyceride levels and preventing endothelial dysfunction, suggesting potential therapeutic utility in cardiovascular and metabolic disorders.
