AFN-12520000 (also known as AFN-1252 or afabicin desphosphono) is a small molecule antibiotic developed for the treatment of acute bacterial skin and skin structure infections (ABSSSI), particularly those caused by *Staphylococcus* species including methicillin-resistant *Staphylococcus aureus* (MRSA)[2][3][4][5]. It acts as a selective inhibitor of enoyl-acyl-carrier protein reductase (FabI/MECR), an essential enzyme in the bacterial fatty acid biosynthesis pathway[4][8]. This mechanism confers high specificity for staphylococci while sparing other bacteria[8]. The drug was originally developed by Affinium Pharmaceuticals and later associated with GSK. Clinical trials have demonstrated its efficacy and safety in Phase 2 studies for various staphylococcal skin infections such as cellulitis, abscesses, wound infections, and burn infections[1][3][5].
