Overview

Afatinib (BIBW 2992) is an oral, irreversible inhibitor of the ErbB family of tyrosine kinases, including epidermal growth factor receptor (EGFR/ErbB1), HER2 (ErbB2), and to a lesser extent HER4 (ErbB4). It blocks signaling pathways involved in the proliferation and survival of cancer cells. Vinorelbine is a semi-synthetic vinca alkaloid that inhibits microtubule assembly, thereby disrupting mitosis and leading to cell death. The combination has been investigated for use in advanced solid tumors, particularly HER2-overexpressing breast cancer and non-small cell lung cancer. Afatinib was developed by Boehringer Ingelheim[1][4][5][6][8].
