Overview

Afatinib + paclitaxel + carboplatin is an experimental combination regimen that includes three antineoplastic agents with distinct mechanisms of action. **Afatinib** is an oral, irreversible inhibitor of the ErbB family of receptors, primarily targeting epidermal growth factor receptor (EGFR), HER2, and HER4, which interferes with cell signaling in cancers driven by these pathways. **Paclitaxel** is a cytotoxic antineoplastic agent that stabilizes microtubules, thereby inhibiting mitosis in rapidly dividing cells. **Carboplatin** is a platinum-containing compound that crosslinks DNA, leading to apoptosis. This combination has been investigated mainly for treatment of patients with advanced solid tumors, including non-small cell lung cancer and other carcinomas, in early-phase clinical trials. The combination aims to enhance antitumor efficacy through synergistic effects between targeted inhibition of growth signaling (afatinib) and cytotoxic chemotherapy (paclitaxel and carboplatin)[1][2][3].
